According to a recent study sponsored by Aschimfarma and conducted during the Arthur D. Little yearly Round Robin, Western CDMOs focused on small molecule API manufacturing invest approximately 11% of sales annually (based on a survey of 16 CDMOs), or nearly 60% of generated earnings before interest, taxes, depreciation, and amortization (EBITDA). Approximately half are focused on maintenance and compliance, with the other half directed toward growth initiatives, such as capacity expansions and the acquisition of new capabilities. Common themes include adapting production infrastructure to meet evolving customer needs, increasing process development capabilities, adding high-containment facilities and equipment, adding particle engineering technologies, exploring continuous processing, and boosting their presence in multiple geographic areas. Some are also adding analytical technologies specifically useful for drug substance development, such as, X-ray powder diffraction (XRPD), thermogravimetric analysis (TGA), and differential scanning calorimetry. Others are investing in pre- formulation development capability, and drug product, with the goal of becoming more full- service providers. The shift toward more highly potent, small-volume APIs that have larger, more complex structures is driving these investment trends, as is the desire for pharma companies to work with fewer service providers and to establish more preferred supplier relationships.

According to one reckoning, China has the greatest number of small molecule API manufacturing facilities, followed by India and the United States (Figure 17).[140] While supply chain security concerns are driving interest in capacity expansion in Western countries, given the large populations in China and India, demand for APIs in these countries will be very large, and any additional capacity added in the United States or Europe is unlikely to change this distribution.
